* INTRODUCTION:
• SpecWhite® TA
is a synthetic derivative of lysine, with a structure partially similar to tyrosinase.
• Its efficacy has been demonstrated through in-vivo studies for functions such as pigmentation removal, skin whitening, and fading dark spots like post-sun exposure hyperpigmentation, brown spots (melasma, freckles, age spots, etc.). It is suitable for sensitive skin, inflamed acne, and also supports post-procedure care such as laser treatments and intense pulsed light (IPL), among others.

* MECHANISM OF ACTION IN MELASMA TREATMENT:
TXA (Tranexamic Acid) prevents UV-induced skin pigmentation by interfering with the structure of plasminogen and blocking its binding to lysine-binding sites on keratinocytes.
• This results in a reduction of free arachidonic acid release, thereby decreasing prostaglandin production — which in turn suppresses tyrosinase activity in melanocytes and inhibits melanin synthesis.
• Additionally, TXA’s effect on angiogenesis through plasmin modulation may also contribute to its efficacy in melasma treatment.
• Inhibition of the Sc-uPA (single-chain urokinase-type plasminogen activator) pathway is another proposed mechanism by which TXA helps reduce hyperpigmentation.
• TXA also partially resembles tyrosine in structure, allowing it to competitively inhibit tyrosinase — the key enzyme in melanin biosynthesis.

* BENEFITS AND CLINICAL STUDIES:
1. WHITENING (IN-VIVO)
Volunteers applied a product containing 0.5% SpecWhite® TA twice daily — in the morning and evening — after facial cleansing for 8 weeks.

👉 Conclusion: After 56 days of use, pigmentation spots under the eyes visibly faded and became nearly unnoticeable. For age spots, the number, total area, and pigmented area ratio significantly decreased after just 14 days. Specifically, the number of age spots was reduced by 12.50% after 56 days compared to the untreated group.

2. WHITENING (IN-VITRO)

Tyrosinase Inhibition Activity of SpecWhite® TA

👉 Conclusion: EC₅₀ refers to the concentration of active ingredient required to inhibit 50% of tyrosinase enzyme activity. An EC₅₀ value of 3.832 mg/mL indicates that SpecWhite® TA exhibits a moderate tyrosinase inhibitory effect.

3. MOISTURIZING (IN-VIVO)
A study was conducted on 12 male and female volunteers using a product containing 2% SpecWhite® TA. Measurements of WCSC (Water Content in the Stratum Corneum) and TEWL (Transepidermal Water Loss) were taken at the following time points: 0h, 1h, 2h, 3h, 4h, 5h, 7h, 24h, and 48h.

👉 Conclusion: SpecWhite® TA at 2% significantly increased water content in the stratum corneum for up to 48 hours and effectively reduced transepidermal water loss, especially within the first 4 hours. The results were noticeably superior compared to purified water, demonstrating outstanding moisturizing efficacy.
